Title: Isolation, identification and characterization of Rhizobium sp

Abstract:

Background :Three Rhizobium strain’s nutritional uptake is same, but the growth rate of BARI RLC107 is higher than other two strains (BARI RCA-220, BARI RVM-307).Different kinds of experiments were set up in both Luria-Bertani (LB) liquid and semi-solid media to observe the growth and maintenance of the strains.

Method: Subculture is required for maintenance the bacterial strain after 21 days. Before starting the subculture program, the flow of the laminar airflow cabinet was cleaned with 70% ethanol for 20 minutes. All the inoculations and aseptic manipulations were carried out in front of the running laminar air flow cabinet. All the dissecting instruments were covered with aluminum foil and autoclaved by steam autoclaving procedures.

Results: strains were isolated and maintained by subculture procedure. Subculture is required after 3 weeks for the maintenance of bacterial strains. Secondly, streaking and plating were observed and counted cell density by plate counting method. Finally, several experiments were set up to optimize growth conditions in LB media: incubated time 48 hours, pH ranging from 7 to 8, temperature ranging from C 0 10 to C 0 40 and peptone as carbon source. Strains grown in the medium supplemented with peptone showed rapid and good performance on growth, and strains grown in glucose showed moderate growth. On the other hand, strains grown in the media containing glycerol and sugar showed poor and slow performance on growth. So, peptone was proved as the best carbon source for these bacterial strains. BARI RLC-107 shows the better performance than the other two strains (BARI RCA-220, BARI RVM-307).

Conclusion: As described before three Rhizobium strains such as BARI RLC-107, BARI RCA-220, BARI RVM-307 collected from Bangladesh Agricultural Research Institute (BARI). The strains belong to the genus Rhizobium but the species is still unknown. Rhizobiums are very important soil bacteria. These strains are naturally able to fix nitrogen which may help us to reduce the utilization of harmful inorganic fertilizer in soil. In this study an attempt was made to optimize the growth characteristics of these three newly isolated strains. The strain grown in the medium containing peptone, pH ranging 7-8 showed the rapid and good performance on growth.
