Abstract:

Molecular biotechnology continues to redefine point-of-care diagnostics by integrating advanced biomolecular recognition with nanotechnology-based detection systems. This study presents the development of a Dual Lateral Flow Immuno Assay (DLFIA) for simultaneous detection of rotavirus and adenovirus antigens in stool samples — two leading causes of acute viral gastroenteritis in children. The biosensor platform combines antibody-antigen molecular specificity with optimized gold nanoparticle conjugation to achieve high analytical sensitivity and rapid signal generation. Distinct capture antibodies were immobilized in separate test zones on a single nitrocellulose membrane, enabling multiplexed detection within 15 minutes. Sample pretreatment using a protein-preserving buffer was formulated to reduce stool viscosity and improve antigen availability without compromising flow dynamics. The DLFIA demonstrated strong signal intensity and stability at room temperature, confirming its suitability for low infrastructure diagnostic environments. This dual-target molecular immunoassay exemplifies the practical application of biotechnology and bioengineering in developing affordable, high performance diagnostic tools, advancing global efforts toward accessible infectious disease surveillance.
