Title : Cloning and expression of Tambaqui recombinant growth hormone (Colossoma macropomum) in Bacillus subtilis

Abstract:

Researches related to the search, cloning and expression of protein-coding genes involved in important fish physiological processes point to a new production scenario in world aquaculture, with the increasing use of techniques and products derived from biotechnology. For its zootechnical importance, the growth hormone (GH) from various species of fish has been successfully produced in heterologous expression systems. Tambaqui (Colossoma macropomum) is a very important species on Brazilian fish farming, whose GH gene (tGH) has been sequenced and deposited in GenBank. This research aimed to express the tGH encoding gene on the prokaryote Bacillus subtilis (PY79). The nucleotide sequence of tGH was cloned into the vector pHT43, and the recombinant plasmid called pHT43-tGH was used for the natural transformation of B. subtilis. Heterologous expression was induced by IPTG and tGH was confirmed recombinant on SDS-PAGE gel and Western blotting. This study demonstrated that tambaqui GH gene may be successfully expressed in B. subtilis, which represents another promising technological breakthrough for growing of this this important Amazon fish species.

Presentation Learning Outcome

  • The research on the Tambaqui growth hormone (tGH) shows the need to speed up the natural process of fish growing, which is beneficial for fish farming, while also providing important information about the bioprocesses involved in production, and the physiological and zootechnical effects when used in fish raised in captivity. Therefore, this work may open new research lines on this subject, promoting interest in research on fish growth and the use of new organisms in biotechnological research.
  • With the results, the research shows that it is possible to create or to optimize Tambaqui GH in different organisms. In addition, it shows a faster, cheaper, efficient solution, by the use of Bacillus subtilis in this process, therefore being of economic interest. The research provides a new product and it encourages new research on other fish species.

Melyssa Oliveira Gomes, graduate student in biological sciences at the Federal Institute of Education, Science and Technology of Amazonas – campus Manaus Centro, Brazil. This research was conducted as Scientific Initiation Project, supervised by Professor Dr. Elson Antônio Sadalla Pinto, and having as a volunteer in the research Aline Silva de Souza, who belongs to the same educational institution.
